Primary Uses & Mechanism
- Indication: Used for HR-positive, HER2-negative advanced or metastatic breast cancer.
- Combination Therapy: It is almost always prescribed alongside hormonal therapies such as letrozole, anastrozole, exemestane, or fulvestrant.
- How it Works: It is a CDK4/6 inhibitor. It blocks specific proteins (cyclin-dependent kinases 4 and 6) that signal cancer cells to grow and divide.

Usage Instructions

- Schedule: Typically taken once daily for 21 consecutive days, followed by a 7-day break.

- With Food: Capsules should be taken with food at approximately the same time each day to ensure consistent absorption.
- Whole Swallow: Do not open, chew, or crush the capsules. If a capsule is broken or cracked, do not use it.

Key Precautions & Side Effects- Blood Monitoring: Regular blood tests are required to check for neutropenia (low white blood cell count), which increases infection risk.
- Interactions: Avoid grapefruit and grapefruit juice, as well as St. John’s Wort, as they can interfere with how the drug is processed.
- Common Side Effects: Fatigue, hair thinning/loss, nausea, diarrhea, decreased appetite, and mouth sores.
- Birth Control: Both men and women must use effective contraception during and for several weeks after treatment (3 weeks for women, 14 weeks for men) due to risks to a developing foetus.
